Robertson, who has played every minute of every game since he made his Sea Eagles debut in round two in 2006, injured his right knee in last week's win over the Gold Coast Titans with the injury worsening over the weekend.
Robertson trained with the squad on Monday with the knee strapped and sore but was ruled out of training on Tuesday morning.
"I'm as shocked as anyone. I remember some discomfort during the match but I didn't think it was serious," Robertson said on the Manly website on Tuesday.
Robertson, who is one of the club's most consistent performers, scored two tries against the Titans and his absence will be a blow for Manly.
The former Canberra speedster's amazing run of consecutive games is the most by any current NRL player.
Retired half-back Jason Taylor holds the record for the most consecutive first grade matches after stringing together 194 games between 1992-2000 for Western Suburbs, North Sydney and the Northern Eagles.
Friday's Lang Park match would have been Robertson's 198th NRL match and his 140th for the Sea Eagles.
